Researchers identify genetic basis of cardiac, craniofacial birth defects

A group of researchers in Israel, the United States and other nations have made important advances in the rapidly-expanding field of "regenerative medicine," outlining for the first time connections in genetic regulation that normally prevent birth defects in heart and facial muscles.

Some of these problems are surprisingly common ? about 1 percent of all people have a congenital heart defect. This basic research will provide a road map to ultimately allow scientists to grow the cell types needed to repair such defects, from stem cells that can be generated from a person's own body.

The findings were published online today in the Proceedings of the National Academy of Sciences.

"Advances in regenerative medicine and developmental biology can now happen because we no longer require human embryos to generate stem cells," said Chrissa Kioussi, a co-author on the study and associate professor in the College of Pharmacy at Oregon State University. "The Nobel Prize this year was awarded to people who discovered how to make stem cells from adult biopsies."

Patient-derived stem cells can in principle be turned into any needed cell type, Kioussi said. The key is understanding the exact regulatory process than tells cells what type they are supposed to turn into, she said, such as a cell on the outside of the left ventricle of the heart.

"Once we understand these genetic controls in sufficient detail, we can not only turn a skin cell into a stem cell, but also turn that stem cell into the type needed for the patient to recover," Kioussi said. "We may eventually be able to grow replacement organs from the patient's cells."

In this study, researchers identified four specific "transcription factor" genes that control processes related to heart and head muscle formation. When there are defects in this process, the result can be death or a range of debilitating problems, from cleft palate to facial malformations and defective heart valves.

"There are about 20,000 genes in the human genome, but only 2,000 of the genes describe transcription factors," Kioussi said. "These transcription factors control the output of genes, the genetic machinery. They collectively determine which of the 20,000 possible molecular machines is actually deployed in each particular cell type."

Scientists have found that these transcription factors don't work alone to define cell types in mammalian development ? they function in small, self-stabilizing combinations of at least two or three.

The process moves rapidly after conception, and within one month most of the cells in the body "know" their cell type, based on the precise combination of transcription factors produced within them. When researchers understand how these stable transcription factor combinations get generated, they will know how to artificially generate these combinations in stem cells to convert them into the needed cell types.

Mammalian embryonic development is a process of self construction, a series of transitions of "temporary" cell types on the way to adult cell types. A fertilized egg is essentially a stem cell with the potential to become any other cell type. At each intermediate stage, the "temporary" cell types become more restricted in what they can become, until they ultimately achieve and maintain the adult type.

"In this work and in regenerative medicine, we care a great deal about all of these steps of cell differentiation," Kioussi said. "If you know all the steps it takes to get from here to there, you can identify what went wrong and find ways to fix it. This is being done already with some disease problems, and this work will move us closer to being able to repair heart and craniofacial defects."

The task is complex, Kioussi said, but very possible. Although there are 100 trillion cells in the human body, there are only about 100 adult cell types. Understanding and influencing the genetic specification of those cell types is possible and will probably revolutionize the treatment of many defects and diseases, Kioussi said.

The lead author was Eldad Tzahor at the Weizman Institute of Science in Israel, and other collaborators were from universities and agencies in the United Kingdom, India and Spain.

Durable orders show business investment still weak

WASHINGTON (AP) ? U.S. companies remained cautious in September and held back on orders for long-lasting manufactured goods that signal investment plans. Weak business investment has contributed to slower economic growth.

The Commerce Department said Thursday that orders for durable goods jumped 9.9 a percent last month. While it was the biggest gain in nearly three years, most of the September gain was driven by a tremendous spike in aircraft orders, which are very volatile and plummeted in the previous month.

When taking out transportation, orders rose just 2 percent. And demand for core capital goods, such as machinery and equipment, were unchanged in September. Core capital goods are considered a proxy for business investment plans. Those orders rose only slightly in August after steep declines in July and June.

Peter Newland, an economist at Barclays, said the report "painted a soft picture of business demand" in the July-September quarter. He said the weakness had prompted him to trim his estimate for economic growth in the quarter from 2 percent down to 1.8 percent.

Durable goods are products expected to last at least three years. Orders can fluctuate sharply from month to month.

Demand is sharply lower this year, which has weakened manufacturing and hampered economic growth. High unemployment and low pay have kept consumers from spending. Businesses have held back on investing in machinery and equipment. And slower global growth has dampened demand for U.S. exports.

Economists are concerned that businesses could pull back further if Congress doesn't reach an agreement to avert steep tax hikes and government spending cuts scheduled to take effect in January.

There is some indication that U.S. factory activity could be improving. A closely watched survey from the Institute for Supply Management, said manufacturing grew in September for the first time in four months. It was buoyed by a jump in new orders.

Americans also boosted their spending at retail businesses in September, buying more cars, appliances and furniture. Stronger consumer spending could help offset declining demand from overseas.

Still, the decline in U.S. manufacturing has slowed job growth. Manufacturing employment dropped by 16,000 in September after falling by 22,000 in August.

The weak job market has been a key topic in this year's presidential election, which is down to its final days. Voters will have one final employment report to consider, which comes out four days before Election Day.

The economy is not growing fast enough to generate much hiring. Growth slowed to a tepid annual rate of 1.3 percent in the April-June quarter, down from 2 percent in the previous quarter. Most economists see growth staying at or below 2 percent in the second half of the year. The Commerce Department will issue its first estimate of growth in the July-September quarter on Friday.

Galactic snack time: Stream of stars slowly being ingested by the Milky Way

ScienceDaily (Oct. 25, 2012) ? Using the Sloan Digital Sky Survey, researchers have discovered a band, or stream, of stars believed to be the remnant of an ancient star cluster slowly being ingested by the Milky Way, Earth's home galaxy.

"The Milky Way is constantly gobbling up small galaxies and star clusters," said Ana Bonaca, a Yale graduate student and lead author of a study forthcoming in Astrophysical Journal Letters. "The more powerful gravity of our Milky Way pulls these objects apart and their stars then become part of the Milky Way itself."

Researchers have previously found evidence of the Milky Way eating up dwarf galaxies. Bonaca argues that the newly found stellar stream is the remnant of a star cluster rather than of a larger galaxy, because the stream is very narrow.

"Our discovery is more of a light snack than a big meal for the Milky Way," says Marla Geha, associate professor of astronomy at Yale and a co-author of the study. "Studying this digestion process in detail is important because it gives us new insight into how all galaxies form and evolve."

The new band of stars, or stellar stream, it is the first of its kind found in the southern Galactic sky, a region that has been hard to examine due to a relative lack of deep-sky imaging there. Deeper imaging enables astronomers to detect fainter stars.

Named the Triangulum stream, the newly discovered stellar stream could also help astronomers reconstruct how the Milky Way's mass is distributed, further revealing its dynamic structure.

Galaxies are believed to form hierarchically through the merger of smaller galaxies and still smaller star clusters. Stellar streams form as they are ripped apart by the gravitational force of galaxies. This process may be the primary way galaxies such as the Milky Way grow in mass, the researchers say.

Triangulum was found by searching a region recently surveyed by the Sloan Digital Sky Survey III (SDSS-III), an international collaboration that is mapping the sky through wide-field photometry.

Bonaca, Geha and co-author Nitya Kallivayalil, a Yale postdoctoral fellow, relied specifically on the survey's Data Release 8, which included information about vast new areas of the southern galactic sky.

President Barack Obama on Thursday threw his support behind ballot measures in Maine, Maryland and Washington state that would legalize same-sex marriage.

Though the president first voiced his general approval for gay marriage in May, he had not previously offered specific endorsements of the three measures.

In each case, the endorsements were issued through the state branches of Obama's re-election campaign.

"While the president does not weigh in on every single ballot measure in every state, the president believes in treating everyone fairly and equally, with dignity and respect," said Paul Bell, the campaign's press secretary in Washington state.

"Washington's same-sex marriage law would treat all Washington couples equally, and that is why the president supports a vote to approve Referendum 74," Bell said.

In Maine, gay-rights supporters put the measure on this year's ballot in hopes of reversing the results of a 2009 referendum in which voters rejected a same-sex marriage law passed by the legislature.

In Maryland and Washington, gay-marriage laws were passed by lawmakers and signed by the governors earlier this year, but opponents collected enough signatures to hold referendums on whether the laws should be upheld or rejected.

Ed Murray, an openly gay state senator in Washington who has been fighting for years to legalize gay marriage, expressed gratitude to Obama.

"When I first began fighting in the legislature for marriage equality ... I would never have dared to dream that a president of the United States would one day step forward at this crucial moment, in the middle of his own close re-election campaign, to offer his support for our efforts," Murray said. "But that is exactly what President Obama has done, and it is an example of his courage and leadership."

Murray said his hope was to soon be able to wed his partner of 21 years, and he thanked Obama for "bringing that dream a little closer to reality."

Chip White of Preserve Marriage Washington, which opposes the gay-marriage law, said he was unsurprised by Obama's action.

"Until May of this year, the president's position was that marriage is the union of one man and one woman," White said. "No one called him a bigot or said he was unfair for holding that position. And Washingtonians who believe in the traditional definition of marriage as one man and one woman are not bigots."

If any of the measures are approved, it would be the first time that a state legalized same-sex marriage through a popular vote. Thus far, all 32 states voting on gay marriage have rebuffed it, while the six states that have legalized it did so through legislation or court orders.

In all three states voting on the issue on Nov. 6, the outcome is expected to be close, though the polls up to now have given an edge to gay-marriage supporters.

Foreclosure wave cost neighbors $2 trillion

The economic impact of the housing bust on homeowners who live near a foreclosed property comes to about $2 trillion, according to a new accounting by a consumer advocacy group.

In a report released Wednesday, the Center for Responsible Lending estimates that more than half of that ?spillover loss? occurred in communities of color, which the study?s authors define as census tracts where more than 50 percent of the residents are Hispanic, African-American?or?otherwise?non-white. Those losses, the authors note, reflect the high concentrations of foreclosures in those neighborhoods.

The study looked only at the indirect impact of foreclosures on properties within one-eighth of a mile of a home in foreclosure. The total loss of home equity since the housing market collapsed is more like $7 trillion, the consumer advocacy group said. That figure ?doesn't?include additional indirect costs.

?Communities with high concentrations of foreclosures lose tax revenue and incur the financial and ?non-financial costs of abandoned properties and neighborhood blight, while homeowners living in close proximity to foreclosures suffer loss of wealth through depreciated home values,? the report?s authors wrote.

Households near a foreclosed property lost or will lose an average $21,077 in wealth or about 7.2 percent of their home value, the report found. In minority neighborhoods, the families lost an average of $37,084 or 13.1 percent of their home value.

To arrive at its estimate, the group combined statistics collected by the government, Mortgage Bankers Association and a private database maintained by Lender Processing Services, a company that provides services to mortgage lenders.

The authors note that over time some of the losses inflicted by the spillover effect of foreclosures will be offset by rising home prices. The estimate covers foreclosures started between 2007 and 2011.

The pace of home new foreclosure filings has been falling this year, falling 7 percent in September compared with the previous month and down 16 percent from September 2011, according to RealtyTrac, a research firm. That was the lowest total since July 2007.

Online Initiative Launches to Aid Low-Income Students

A new initiative in online learning intending to help low-income adults achieve an associate?s degree is set to launch in Colorado and then California before expanding to selected cities nationwide.?

MyCollege Foundation is partnering with Mount St. Mary?s College in Los Angeles in this program that aims to give students who are strong on grit and drive the chance to gain the skills and confidence to earn a two-year degree, without taking on extensive debt before advancing toward a bachelor?s degree.

Partly funded by a?$3 million grant?by the Bill & Melinda Gates Foundation, Portmont College at Mount St. Mary?s plans to offer four fields of study: business administration, computer science, liberal arts, and pre-health science. The concept is the brainchild of Portmount President?Srikant Vasan, developed in part while he served as entrepreneur-in-residence at the Gates Foundation.?

The Portmont program starts with face-to-face instruction with professors during kick-off orientation, followed by online coursework. It will also have support services to ensure that students graduate. ?

Students can transfer their credits to four-year institutions to complete a bachelor?s. Portmont will launch in Denver, followed by San Francisco. Classes in Denver start on March 4.

While the $5,240 per year price tag is higher than the average ($2,200) for Colorado community colleges, the cost of attending Portmont is significantly lower than studying at Mount St. Mary?s, an independent, Catholic liberal-arts women?s college. Tuition and fees there exceed $31,000 per year.

National figures show that student-loan debt has reached historic highs. About one in five households has debt tied to obtaining a degree. Compared with other demographic groups, a smaller percentage?(31 percent)?of Hispanics borrowed money to pay for school; the figures are 37 percent for whites and 35 percent for African-Americans, a national survey on?How America Pays for College?by the Sallie Mae Fund.

Still, the amount of money that students have borrowed to pay for college has ballooned. A?new report?indicates that 2011 college graduates had an average of $26,600 in student loans, about 5 percent higher than students who completed a degree program the previous year. Only six years ago, students were graduating with approximately?$19,600?in student loans.

  • ?Success Launch Process,? which goes beyond the cognitive topics typically addressed by college success courses and uses a series of four courses to develop intrapersonal and interpersonal skills correlated to work and life success.
  • Personal support network of success coaches, faculty, and peers so that each student benefits from tailored, proactive interventions using student-specific academic and behavioral data.
  • Technology platform offering visibility into key progress measures to students, faculty, and employers ? includes holistic performance monitoring, historical reporting, and predictive analytics ? enabling proactive, tailored interventions.

Gay GOP group hesitantly backs Romney

A prominent gay Republican group is hesitantly backing Mitt Romney for president, calling it the right decision for the nation even as it slams his opposition to gay marriage and civil unions.

Log Cabin Republicans says it's supporting Romney to put America and the economy first, but is only offering him a "qualified endorsement." The group says it will focus its efforts instead on House and Senate candidates.

In its endorsement, Log Cabin Republicans says Romney might not be the candidate for voters who prioritize gay and lesbian issues. The group has long been critical of Romney and says it will continue challenging his support for a federal amendment banning gay marriage.

The group is also condemning aspects of the GOP platform that, quote, "work to exclude our families."

2 killed in overnight clashes in Lebanon

BEIRUT (AP) ? Clashes in Beirut and other parts of Lebanon killed at least two people and left more than a dozen wounded overnight as fears grow that Syria's civil war is spilling over into its smaller neighbor, security officials and state-run media said Monday.

The latest spasms of violence came amid rising tension between Lebanese groups that support and oppose the regime of Syrian President Bashar Assad and in the wake of the assassination last week of a top anti-Syrian intelligence official in Lebanon.

Most of Lebanon's Sunnis have backed Syria's mainly Sunni rebels, while Lebanese Shiites tend to back Assad. The Syrian president, like many who dominate his regime, is a member of the Alawite sect, an offshoot of Shiite Islam.

Lebanese Brig. Gen. Wissam al-Hassan, who was killed in a car bomb in an east Beirut neighborhood on Friday, was a Sunni who challenged Syria and Hezbollah.

His assassination has threatened to shatter Lebanon's fragile political balance. Many politicians have blamed Syria for the killing and angry protesters tried to storm the government palace after al-Hassan's funeral Sunday but were pushed back by troops who opened fire in the air and fired tear gas.

Overnight, Sunni and Shiite gunmen clashed in two Beirut neighborhoods and officials also reported heavy clashes late Sunday and early Monday in the northern city of Tripoli and towns between Beirut and the southern city of Sidon.

The officials, speaking on condition of anonymity because they were not authorized to talk to the media, said a man was killed in shooting in the Wadi Zayneh area north of Sidon and another person died in the Tripoli clashes. The officials said the clashes wounded at least six people in Beirut and 10 in Tripoli.

An Associated Press photographer saw dozens of gunmen roaming the streets Monday of Beirut's predominantly Sunni neighborhood of Tariq Jadideh, where much of the fighting had taken place. Local Sunni leaders were calling the gunmen by telephone urging them to pull out of the streets.

In some roads around Tariq Jadideh, masked Sunni gunmen set up checkpoints, stopping cars and asking people about their destination and where they were coming from.

A woman who lives in the neighborhood said the fighting began shortly after midnight and lasted until sunrise.

"We couldn't sleep because of the shooting. There were also some booms," she said, referring to rocket-propelled grenades. She asked that her name not be mused for fear of reprisals.

In Tripoli, residents said scores of soldiers deployed around the city in an attempt to bring back calm. The military also set up checkpoints, searched cars and asked people for identity cards.

(Let?s not forget these people also have three young children!)

The Zavros family live and work in this sprawling four bedroom property, where they?ve been based for just over five years. ?The home is actually in Chandler, around 30 minutes out of central Brisbane, on a generous 8 acre block which also incorporates a purpose built studio for Michael, swimming pool, tennis court and chicken house. ?Whilst the Zavros? home itself is still waiting for the ?big? renovation, extensive work has been done to two large industrial sheds on the grounds, to create Michael?s studio and a self contained guest house.

It?s clear Michael and Alison are avid collectors of many special things ? despite the presence of three very energetic young kids, their home certainly feels museum-like, full of valuable treasures! ??It?s an unusual mix of antiques and artworks? says Michael. ??We have a Bill Henson work that we love, we seem to have a penchant for chairs ? more chairs than people to sit in them. We also love our many Chesterfields, probably because they are all about sitting and relaxing, drinking wine and watching good telly. They are our sitting happy places!?

Not surprisingly, given their backgrounds, many of Michael and Alison?s dearest friends are artists, and most of the artworks you see here are swaps or gifts, and hold great sentimental value. ??Curiously, when gathered together, they all seem to be about portraiture? says Michael of the collection. ?Highlights include?works by Shaun Gladwell, Bill Henson and Susan Norrie.

Michael and Alison love the serenity of living outside of the city on a generous block ? ?It?s good to not hear your neighbours sneeze? says Michael. ?We love that the kids can throw on their boots and just be outdoors all day. ?At night wallabies bound about outside the bedroom and we have seen several koalas up close. Nature abounds!?

Is Adele officially a mom? According to multiple reports out of the UK, the notoriously private singer has given birth to a baby boy.

A Note From Girl Scouts of Alaska's New CEO | alaskapublic.org

This is my second week on the job as Girl Scouts of Alaska?s new Chief Executive Officer. I?m excited about the coming year and looking forward to meeting as many of you as possible.

I?m making arrangements to visit many of the communities Girl Scouts of Alaska serves. It?s important that I meet and talk with as many of you as possible about what?s working and what you might like to see improve, so that we can best support you in delivering the Girl Scout Leadership Experience. Stay tuned for more details about where I?ll be visiting and when.

In the meantime, I thought I?d share with you a little bit about myself and why I wanted to be CEO of Girl Scouts of Alaska.

My family called Fairbanks home when I was a teenager. I spent summers working on the first all-women?s fire-fighting crew for the Bureau of Land Management (BLM), interning for Alaska?s congressman, and working on a research project for the Alaska Native Corporations at the University of Alaska?s Institute for Social and Economic Research.

During the academic year I earned a degree in economics at Princeton. Following graduation, I worked in Washington at the Congressional Budget Office and then headed to Harvard Business School where I earned a Masters in Business Administration. While I was in business school, the Rhodes Scholarship opened applications to women. I attended Oxford as Alaska?s first female Rhodes Scholar where I earned a doctorate in economics, writing my thesis about bidding theory and oil and gas lease auctions.

Since then, I have focused on business, working for a management consulting firm, a large bank, and a small mergers and acquisitions firm. Throughout those years, I always volunteered to help children?s after-school programs ? mostly children?s sports and recreation programs ? grow and flourish.

participating in sports and recreation. With the help of
volunteer Edith Johnston, Girl Scouting?s first basketball
tournament took place in Savannah, Georgia in 1912.

I understand what it is to be a volunteer and what it takes to offer successful after-school programs to children. Our girls deserve the very best we can offer and I look forward to working with you to bring an outstanding range of experiences to Alaska?s girls.

I feel privileged to have this opportunity to work with you to make a difference for girls in Alaska. As you know, the programs and services delivered by Girl Scouts wouldn?t be possible without our 1,500 volunteers to serve as mentors, role models, chaperones, and friends to girls.

Collectively, volunteers contribute thousands of service hours to Girl Scouts.

Just as we always ask the girls what they want in order to keep our programs relevant and interesting to girls, I want to keep an open dialogue with our volunteers, to ensure that you, too, are engaged and have the resources to support the incredible experience of sharing your knowledge and wisdom with girls.

I?m interested in hearing from you. In the coming weeks and months, start looking for my Facebook posts on the Girl Scouts of Alaska Facebook page, and on our blog. Feel free to comment and respond directly to me. I?ll do my best to answer your questions and I always welcome your input and ideas.
